This publication describes the discovery of the yeast and mammalian pro-protein and pro-hormone convertases kexin (Kex2), furin (PACE), PC1(PC3) and PC2. The most up-to-date properties, neuroanatomical distribution, gene regulation and cleavage specificity of each enzyme are described together with a definition of the role of the prosegment in the autoactivation of bacterial subtilisins. Furthermore, among the many exclusive features of this publication is a first description of the recently discovered subtilisin-like convertases called PACE4, PC5 and PC6.
