This text presents the latest techniques in the study of biological membranes and transport processes. This area of investigation has grown due to technical advances in four important areas: electrophysical recording of transport events; intracellular ion-sensitive dyes; optical imaging; and biochemical and molecular biological approaches to the study of transport systems. Descriptions are given for both the basic theoretical and practical aspects of these methodologies, as well as their applications to a variety of membranes and transporters in isolated cells, epithelial cell layers and artificial membrane systems. It examines the use of patch-clamp, and noise and impedance analysis to study the kinetic properties of individual ion channels in living and in vitro cells, as well as in isolated biological and artificial membranes. The use of ion-selective intracellular micro-electrodes is explored and there is a discussion of special problems and techniques for measuring intracellular pH and Ca2+ activity.
