Molecular biology experienced a revolution in the 1960s and 70s. Most of the great strides forward during this period were accomplished using E. Coli as an experimental model - a unicellular bacterium with no nucleus in its cell. However, in order to have any hope of relating molecular biology to man in terms of either pure understanding or applications, it was necessary to come to grips with multicellular organisms incorporating a nucleus within their cellular membranes. The simplest (and least expensive) such model to culture and study is Saccharomyces Cerevisiae (Baker's Yeast) - and hence the vast amount of work being done all over the world on this organism. In this manual, the authors cover all the laboratory methods used to culture, purify, transform, translate, splice and express the genomes of yeast.
